Biochemical and molecular investigations on qualitative and quantitative Hb polymorphism in the river buffalo (Bubalus bubalis L.) population reared in Southern Italy Genet. Mol. Biol.
Iorio,Mario; Vincenti,Donatella; Annunziata,Mario; Rullo,Rosario; Bonamassa,Raffaele; Di Luccia,Aldo; Pieragostini,Elisa.
On 398 river buffalo samples, randomly collected in distinct breeding areas of the Campania region, high-resolution analytical systems were used to identify both qualitative and quantitative variations of the Hb phenotype. Polyacrylamide gel isoelectric focusing and HPLC were used to determine the ratio between HBA1 and HBA2 globin chains; restriction endonuclease analysis was performed to assess whether quantitative variations in Hb bands were related to an unusual number of a-globin genes. In the two buffalo subpopulations, allele frequencies of the alpha and beta globin systems were calculated, and F statistics (FIS, FIT and FST) were estimated as parameters of genetic diversity. The results suggest that: i) as shown by RFLP analysis, only a couple of...

Characterization and lytic activity of Pseudomonas fluorescens phages from sewage BJM
Radhakrishnan,Ananthi; Ananthasubramanian,M.
Pseudomonas fluorescens phages from sewage were tested against P. fluorescens isolates of soil and sewage. The phages were characterized as to host range, morphology, structural proteins and genome fingerprint. Of the seven phages isolated, one was found to be abundant in sewage (5.9×10(7) pfu/mL), having broad host range, and distinct protein and DNA profile when compared to the other six phages. DNA restriction and protein profiles of the phages and their morphology indicate the diversity in the sewage environment. None of the isolates from the rhizosphere regions of various cultivated soils were susceptible to phages isolated from sewage.

Soares-Ramos,Juliana R.L.; Ramos,Humberto J.O.; Cruz,Leonardo M.; Chubatsu,Leda S.; Pedrosa,Fábio O.; Rigo,Liu U.; Souza,Emanuel M..
Herbaspirillum spp. are endophytic diazotrophic bacteria associated with important agricultural crops. In this work, we analyzed six strains of H. seropedicae (Z78, M2, ZA69, ZA95, Z152, and Z67) and one strain of H. rubrisubalbicans (M4) by restriction fragment length polymorphism (RFLP) using HindIII or DraI restriction endonucleases, random amplified polymorphic DNA (RAPD), and partial sequencing of 16S rDNA. The results of these analyses ascribed the strains studied to three distinct groups: group I, consisting of M2 and M4; group II, of ZA69; and group III, of ZA95, Z78, Z67, and Z152. RAPD fingerprinting showed a higher variability than the other methods, and each strain had a unique electrophoretic pattern with five of the six primers used....

Comparison of multiple genotyping methods for the identification of the cancer predisposing founder mutation p.R337H in TP53 Genet. Mol. Biol.
Fitarelli-Kiehl,Mariana; Macedo,Gabriel S.; Schlatter,Rosane Paixão; Koehler-Santos,Patricia; Matte,Ursula da Silveira; Ashton-Prolla,Patricia; Giacomazzi,Juliana.
Abstract Germline mutations in the TP53 gene are associated with Li-Fraumeni and Li-Fraumeni-Like Syndromes, characterized by increased predisposition to early-onset cancers. In Brazil, the prevalence of the TP53-p.R337H germline mutation is exceedingly high in the general population and in cancer-affected patients, probably as result of a founder effect. Several genotyping methods are used for the molecular diagnosis of LFS/LFL, however Sanger sequencing is still considered the gold standard. We compared performance, cost and turnaround time of Sanger sequencing, PCR-RFLP, TaqMan-PCR and HRM in the p.R337H genotyping. The performance was determined by analysis of 95 genomic DNA samples and results were 100% concordant for all methods. Sequencing was the...

Evaluation of the protective capacity of new mild Citrus tristeza virus (CTV) isolates selected for a preimmunization program Scientia Agricola
Zanutto,Carlos Alexandre; Corazza,Maria Júlia; Nunes,William Mário de Carvalho; Müller,Gerd Walter.
The use of tolerant rootstocks and preimmunization has satisfactorily controlled losses associated with the Citrus tristeza virus (CTV). Several researchers have shown that CTV mild isolates that are selected in the same region where they are used are superior to isolates obtained from other areas. Thus, budwoods of 20 outstanding citrus trees were collected in north and northwestern Paraná state (Brazil) citrus-producing areas and established to be used in a preimmunization program. These budwoods were tested to evaluate the potential protection of the inherently present viral complex. Based on biological indexing and molecular characterization of the capsid protein gene by RFLP (restriction fragment length polymorphism, which indicated that the plants...

Genetic structure of Brazilian populations of white mouth croaker Micropogonias furnieri (Perciformes : Sciaenidae) BABT
Puchnick-Legat,Angela; Levy,José Alberto.
Mitochondrial (mt) DNA population structure was examined by using restriction fragment length polymorphism analysis (RFLP) among 149 white mouth croakers (Micropogonias furnieri) sampled from six locations in the Brazilian coast. Heterogeneity tests revealed no differentiation in mtDNA haplotype frequencies within the region between 23°S and 34°S (P=0.263), but significant heterogeneity was detected between north and south of 23°S (P<0.003). Analysis of molecular variance (AMOVA) indicated a low F ST value (0.008, P=0.180) among south-central localities (23-34°S), but a significant level of population subdivision (F ST = 0.322, P< 0.003) between north and south-central localities. UPGMA analysis of mtDNA sequence divergence revealed differentiation...
